การตรวจสอบความถูกต้องของการเพิ่มปริมาณดีเอ็นเอชุดซ้ำของเครื่องหมายดีเอ็นเอ จำนวน 16 และ 22 ตำแหน่งในการพิสูจน์เอกลักษณ์บุคคล

Abstract

ปัจจุบันงานด้านนิติพันธุศาสตร์มีการประยุกต์ใช้ในกระบวนการยุติธรรม การพิสูจน์เอกลักษณ์บุคคล ตลอดจนการพิสูจน์ความสัมพันธ์บิดา-มารดา-บุตร ดังนั้นนักนิติพันธุศาสตร์ในห้องปฏิบัติการ บริการ จึงมีความพยายามที่จะการปรับปรุงกระบวนการบริการด้านการตรวจวิเคราะห์ดีเอ็นเอ ให้มีความไว ความถูกต้อง ให้รายงานผลการตรวจวิเคราะห์ที่น่าเชื่อถือ และประหยัดค่าใช้ง่าย งานวิจัยนี้ มีวัตถุประสงค์เพื่อศึกษาคุณภาพของลายพิมพ์ดีเอ็นเอที่ได้จากการลดปริมาตร น้ำยาสำเร็จรูป ในการเพิ่มปริมาณดีเอ็นเอในหลอดทดลอง โดยใช้เครื่องหมายพันธุกรรมบนออโทโซมจำนวน 16 และ 22 ตำแหน่ง จากเซลล์เยื่อบุกระพุ้งแก้ม ด้วยชุดน้ำยาสำเร็จรูป Investigator IDplex GO และ น้ำ1 Investigator 24plex QS พบว่า สามารถลดปริมาณน้ำยาสำเร็จรูปเหลือร้อยละ 50 และ 25 ตามลำดับ โดยที่ให้ผลลายพิมพ์ดีเอ็นเอที่สมบูรณ์และสามารถนำมาใช้ในการตรวจพิสูจน์เอกลักษณ์บุคคลในทางนิติวิทยาศาสตร์ได้ และยังสามารถช่วยลดเวลาและต้นทุนค่าใช้จ่ายของห้องปฏิบัติการ

Recently the knowledge and skill of forensic genetics were applied to justice system, personal identification and paternity test. Therefore, forensic scientists in service laboratories try to develop laboratory process to increase the sensitivity and accuracy, to generate reliablc results and to reduce cost. The purpose of this work is to sturdy the quality of DNA profiles using reduced volume of commercial kits in 16 and 22 autosomal short tandem repeat (STR) markers from buccal cells using Investigator IDplex GO and Investigator 24plex QS kits, respectively. The results showed that complete DNA profiles of 16 and 22 STR markers can be obtained by reducing the kits to 50 and 25 %, respectively. The amplification volume reduction can be used to investigate DNA profiles in personal identification and can save the cost in the routine service laboratory
